In many ways, DNA profiling technology is very similar to the conventional techniques used for forensic identification. As with, for example, blood grouping techniques, the molecular characteristics of the scene of crime sample may be determined and compared with those of the scene of reference samples from suspects and victim. If the molecular characteristics of the crime sample and the suspect are different, then they cannot be from the same person, whereas if they match, then the possibility remains that they may be from a single source.

DNA fingerprinting , also called DNA typing, DNA profiling, genetic fingerprinting, genotyping, or identity testing , in genetics , method of isolating and identifying variable elements within the base-pair sequence of DNA deoxyribonucleic acid. The technique was developed in by British geneticist Alec Jeffreys, after he noticed that certain sequences of highly variable DNA known as minisatellites , which do not contribute to the functions of genes , are repeated within genes. Jeffreys recognized that each individual has a unique pattern of minisatellites the only exceptions being multiple individuals from a single zygote , such as identical twins. It is also used to identify inherited genetic diseases and can be used to identify genetic matches between tissue donors and recipients. DNA fingerprinting is also a valuable tool for confirming pedigree in animals, such as purebred dogs and racehorses.

The blood is treated with a series of chemicals until pure DNA emerges as a white solid. The DNA is stored, dissolved in essentially water, in a small plastic tube and kept in a fridge until ready for the next stage.

In criminal investigations, the DNA fingerprint of a suspect's blood or other body material is compared to that of the evidence from the crime scene to see how closely they match. The technique can also be used to establish paternity. First developed in by Alec Jeffreys, a British professor of genetics at the Univ. All states have established DNA fingerprint databases and require the collection of DNA from convicted felons, and the Federal Bureau of Investigation has instituted a national DNA fingerprint database linking those of the states and including DNA collected in connection with federal offenses. DNA fingerprinting is generally regarded as a reliable forensic tool when properly done, but some scientists have called for wider sampling of human DNA to insure that the segments analyzed are indeed highly variable for all ethnic and racial groups. It is possible to create false genetic samples and use them to misdirect forensic investigators, but if those samples have been produced using gene amplification techniques they can be distinguished from normal DNA evidence.

To ensure correct collection, storage, and use of biological material, a biobank requires standard operating procedures, not only to guarantee the integrity of the sample, but also for the traceability and uniqueness of the same sample and to avoid potential contamination.

In AFLP analysis, bacterial genomic DNA is digested with restriction enzymes, ligated to adapters, and a subset of DNA fragments are amplified using primers containing 16 adapter defined sequences with one additional arbitrary nucleotide. Polymorphisms of different Escherichia coli strains or Agrobacterium tumefaciens strains were demonstrated as distinct, unique bands in a denaturing sequencing gel using AFLP. DNA fingerprinting techniques such as restriction fragment length polymorphism RFLP and random primer polymorphism amplification detection RAPD have been described as powerful molecular typing methods for microorganisms 1. AFLP has been used to establish genetic linkage maps and to localize disease resistant genes 3 , 4. There are three major steps in the AFLP procedure: i restriction endonuclease digestion of genomic DNA and the ligation of specific adapters; ii amplification of the restriction fragments by PCR using primer pairs containing common sequences of the adapter and one to three arbitrary nucleotides; iii analysis of the amplified fragments using gel electrophoresis. The combination of different restriction enzymes and the choice of selective nucleotides in the primers for PCR make AFLP a useful new system for molecular typing of microorganisms. Genomic DNA was isolated from bacterial cultures using lysozyme and detergent 1.

DNA Fingerprinting: Approaches and Applications

This paper aims to describe the basics to understand the various methods of DNA fingerprinting and its. applications in forensic science.

Genetic identity testing involves identifying the patterns of genetic material that are unique to almost every individual.
